Unlock Cooperative Mode in ALL Missions
To unlock Cooperative mode in all missions, including the sub-missions (Wesker's missions), simply complete "The Fourth Survivor" sub-mission on any difficulty. An option will appear on the right when selecting characters that looks like two pistols and says "begin play with 2 players". Both players play as the same character, there are just two aiming reticles on the screen to play with.

Comment: thanks.. is "The Fourth Survivor" Hunks mission right? if so ive done that and played all the missions and sub-missions and completed the game and i have the special stage aswel but still when i go to player select i dont have the option of playing in 2 player mode. what am i doing wrong?

Comment: Just completed the game with all Sub-missions (On Easy).
I just hooked up the 2nd Wiimote and then I could play with Two player co-op on the Sub-Missions (the ones with only one character, Wesker, Rebecca, Ada, Hunk). 
The other levels where you can choose between two Characters, (Train Derailment, Mansion Incident etc.), you can play Two player co-op from the beginning of the game, just plug in a second Wiimote.
